HPA Specific Antigen 1-6,15,21W Genotyping Kit (Fluorescence PCR Method)

【Test principle】This kit uses real-time fluorescence PCR technology to determine the HPA genotype. This method uses primers to amplify DNA fragments containing polymorphic sites, while designing different fluorescent probes based on the differences base of the antithetic antigens. By detecting the intensity of the activated fluorescent probe (S), it determines whether there is a corresponding base sequence, and thus specifies the allele genotype. Each PCR reaction mixture includes a pair of primers and two different fluorescent probes. By comparing the intensity of the fluorescent probes, the base sequence is determined. By detecting 8 responses each time, HPA genotypes 1-6, 15, and 21 can be detected. 96 well plate test can be conducted for 12 people.

Test significance

1.HPA is an inherent antigen of platelets and has polymorphism, which causes single amino acid variation in corresponding positions.

2. If the platelet homologous antigen is not matched, it can lead to platelet homologous antibodies in the recipient, resulting in alloimmune thrombocytopenia, purpura after blood transfusion, and ineffective platelet transfusion.

3. The establishment of HPA gene donor bank to provide patients with platelet preparations of the same type can ensure the safety and effectiveness of platelet transfusion to the greatest extent, which is of great significance for clinical blood transfusion.
